The Church of St. James the Greater stands at Dorpsstraat 41. The basilical cruciform church has a three-sided closed choir and an integrated tower with a spire. Theo Molkenboer designed the building, which was constructed between 1857 and 1859. The church is considered a provincial monument and is therefore a prominent part of the village's history.

The Church of St. James the Greater, restored in 1978, contains a richly carved high altar, a pulpit, Stations of the Cross and a communion rail dating from around 1860. It also has a Ypma organ from 1875. At Dorpsstraat 39 there is also a group of buildings considered a provincial monument and associated with the functions of church and vicarage.

Kerkbuurt can be recognised as Tuitjenhorn's original main centre. Thanks to the adjacent park, this distinctive small settlement quarter is still easy to understand. The village's origins also shape the way its structure is viewed: Tuitjenhorn originally developed as an elongated ridge made up of dwelling mounds, or so-called “walls”.
